The global overpopulation of stray cats and dogs, as well as certain wild and invasive species, poses major challenges for animal welfare, public health, and ecosystem conservation. Surgical sterilization, relocation, culling, and immunocontraception are the primary methods currently used to control animal populations. However, depending on the target species, these approaches may be difficult to implement on a large scale, lack long-term effectiveness, or raise important ethical concerns.

Our laboratory develops long-lasting injectable contraceptive strategies for animal population control.

A major focus of our research is gene therapy. This approach consists of delivering, through a single injection, a therapeutic gene that enables the patient’s own cells to produce a therapeutic protein for several years. Already used in human medicine to treat genetic diseases once considered incurable, gene therapy is now creating new opportunities in veterinary medicine.

Our non-surgical sterilization strategy uses gene therapy to induce the animal’s skeletal muscles to produce Anti-Müllerian Hormone (AMH). In adult females, AMH is normally produced exclusively by the ovaries and acts as a negative regulator of ovarian follicle growth, the structures that contain oocytes. Following a single injection, the treated animal’s muscles continuously produce high circulating levels of AMH, suppressing ovarian follicle development for several years. We have demonstrated that this approach provides long-term prevention of ovulation and fertility in both mice and domestic cats.

In the short term, our laboratory aims to optimize the feline contraceptive and further characterize the effects of sustained high AMH levels on the female reproductive system. Our research combines cell and follicle culture models with in vivo studies to investigate the mechanism of action, safety, and contraceptive efficacy of our therapies. In the longer term, we aim to develop injectable contraceptives for additional animal species and explore new gene therapy–based strategies for reproductive management.

Our laboratory enthusiastically welcomes undergraduate and graduate students, as well as research trainees, interested in projects at the interface of animal reproduction, ovarian physiology, gene therapy, molecular biology, and translational veterinary medicine.

Microtubules are part of the cellular cytoskeleton that orchestrate a variety of processes, including cell division and intracellular trafficking. Although the term “cytoskeleton” suggests a static framework, microtubules are highly dynamic structures that continuously undergo cycles of growth and shrinkage within the cell’s cytoplasm. This dynamic behavior is regulated by microtubule-associated proteins (MAPs), motor proteins, and tubulin post-translational modifications (PTMs). Disruption of microtubule dynamics can impair essential cellular processes such as cell division and migration, thereby contributing to a range of disorders, including cancer, developmental abnormalities, and infertility. Our lab employs genetics, cell biology, live-cell microscopy, and proteomics to investigate how MAPs and PTMs regulate microtubule dynamics in somatic cells, oocytes, and embryos. We are also exploring how specific reproductive biology techniques—such as cryopreservation—affect the microtubule cytoskeleton, and how modifying these protocols can help mitigate negative impacts. Ultimately, our goal is to uncover how altered microtubule dynamics contribute to diseases like cancer, developmental disorders, and infertility, and to apply our understanding of microtubule biology to enhance assisted reproductive technologies.

Dr. Talbi’s research aims to understand how sex hormones and sex differences shape brain function and regulate physiological balance. Specifically, her work investigates how disruptions in energy balance, from undernutrition to obesity, impair reproductive function, and how reproductive hormones, in turn, influence metabolic homeostasis. Moreover, her research is motivated by advancing women’s health through the identification of brain pathways linking reproductive dysfunction with metabolic and hormonal disturbances, and the consequences of hormonal imbalance and endocrine decline across the lifespan.

My research focuses on comparative reproductive pathology, with a particular interest in ovarian infertility, cystic ovarian pathologies across species, as well as the tumor biology of the ovary and the mammary gland. My research program is structured around two main axes:

  1. To better understand the mechanisms underlying reproductive diseases and to develop innovative biological models that bridge veterinary and human medicine.
  2. To promote innovation in the diagnosis of ovarian and mammary tumors, as well as the identification of relevant cellular and molecular markers to improve the detection and understanding of these pathologies.

Eukaryotic genomes are filled with tandem repeats that form long arrays, known as satellite DNA, which currently have no well-understood functions. The research in my laboratory aims to understand the functional impact of satellite DNA activities on male and female reproduction, using Drosophila as a model organism. Particularly, we have three main goals. First, we are interested in how instability of satellite DNA located near the centromere of chromosomes could contribute to chromosome segregation errors. We have developed a system that will allow us to determine factors at the DNA sequence and protein-binding level that cause varying rates of nondisjunction in Drosophila meiosis. Second, we are interested in how repeats present in the introns of fertility genes affects their expression in sperm development. Here we study two closely related species that have a 3-fold difference in the size of the Y chromosome, caused by the expansion of a single repeat family. Finally, we are interested in the evolution and possible functions of long noncoding RNAs produced from satellite DNA that are expressed in the germline.

Dr. Silva’s expertise is centered on the neuroendocrine control of reproductive function and socio-sexual behaviors in mammals. Specifically, my research has provided insights into how hypothalamic and sensory neurocircuits regulate the secretion of gonadotropin-releasing hormone (GnRH). He has also extensively studied how the brain plays a crucial pathogenic role in the manifestation of polycystic ovary syndrome (PCOS), an endocrine disorder that significantly impacts female fertility.

I study the microbial mechanisms leading to pathologies or health in women. An optimal microbial community within the vaginal tract correlates with greater reproductive outcomes (better fertility, pregnancy to term, protection against microbial infections) and this correlation is getting more attention recently, however little is known about the real importance of these microbes for the reproductive system. In long-term, a better understanding of the vaginal and uterine microbes will provide new avenues of prevention and therapies to favor the optimal microbiota and improve reproductive health.

The research group is interested in understanding the cellular and molecular functions of sex steroid hormone receptors. In particular, the team is studying the action of androgens and estrogens in the prostate and mammary gland, as well as the impact of endocrine disruptors on these hormonal signalings, all using rodent models and different types of human organoids. Particular attention is paid to understanding how these hormonal signals reprogram cell metabolism, for example to support lactation by the mammary gland.

My laboratory focuses on the role of inflammatory and gestational cytokines in the regulation of inflammation and energy metabolism in cancer development and gestational diseases. More specifically, our studies aim to understand how certain cytokines and other synthetic molecules with anti-inflammatory action modulate the function and metabolism of macrophages, tumor cells and embryonic cells. This will enable us to discover the biological markers to target in a preventive or curative therapy approach for these diseases.

Our lab’s focus has been deciphering the mechanisms and biological roles of epigenetic reprogramming for pluripotency and germ cell/oocyte development. We specifically study the processes of X-chromosome in- and reactivation, classical models of epigenetic regulation, by developing tailored mouse and human iPSC-reprogramming and organoid systems of germ cell and oocyte development. We thereby made fundamental discoveries on the interplay between 3D-genome structure, epigenetics and transcription and the importance of X-chromosome remodeling for meiosis and oogenesis. Furthermore, together with the clinic, we studied by single-cell RNA-Seq the impact of aging on the human oocyte transcriptome and derived human iPSC-lines of different cell origin and tested their capacity for germ cell differentiation.
Our lab’s future goal is to further advance our knowledge on the epigenetic reprogramming events during germ cell development and thereby to contribute to the development of better in vitro organoid models of mouse and human oocyte development, which recapitulate the in vivo situation more faithfully. These in vitro models will open the reproduction field to in-depth mechanistic studies, and will also facilitate patient-specific disease modeling, offering invaluable insights into human fertility disorders. Furthermore, such in vitro systems could provide versatile platforms for testing fertility or contraceptive drugs and for assessing the impact of environmental toxins, chemicals, or drugs on the reproductive system.
